We Have Obtained a Patent for the Technology Behind “PtWAVE,” a System for Estimating Mutation Introduction Efficiency in Genome Editing: 2026-09-14

PtBio is pleased to announce that we have obtained a patent for an invention entitled “Method for Estimating the Efficiency of Mutation Introduction Using a Genome Editing Tool.”

This is the first patent that PtBio has independently filed and successfully obtained (patent granted). Our mutation introduction efficiency estimation system incorporating this patented technology is PtWAVE.


■ Background and Key Features of the Technology

In the development of improved varieties and useful organisms using genome editing technologies, it is extremely important to determine how efficiently and at what frequency a designed genome editing tool can introduce mutations at the intended genomic location. This is essential for shortening development timelines and improving the probability of success.

The patented technology covers an original method for calculating and estimating the efficiency of mutation introduction by genome editing tools.

Conventional estimation methods, such as Tracking of Indels by Decomposition (TIDE) analysis, have a limitation in that their accuracy decreases when large deletions exceeding 40 base pairs occur. Our technology addresses this challenge by generating multiple in silico sequences, including sequences with large deletions of more than 40 base pairs, based on sequence information from the target region. It then repeatedly constructs and evaluates regression models combining these virtual sequences to estimate the most plausible mutation alleles.

By using both the coefficient of determination (R²) as an indicator of accuracy and the Bayesian Information Criterion (BIC) as an indicator of model completeness, this method enables highly reliable estimation even for complex mutation patterns. This helps prevent large deletions from being overlooked and enables more efficient optimization of genome editing conditions.




We anticipate that this technology will be useful in the following applications:

  • Rapidly narrowing down optimal genome editing conditions

    The system can dramatically reduce the time required to determine the optimal conditions for genome editing, including the type of editing tool and culture environment.

  • More reliably obtaining the desired cells

    By estimating the frequency of mutation allele patterns, including large deletions, the system enables quantitative assessment of how readily the desired genetic mutations can be obtained.
